I mean, kind of. She was originally (and continued to be for most of her existence as a character) a SHIELD agent. Then she had the title of Madame Hydra briefly, but in the course of that storyline it was revealed (retconned) that she had been a Russian sleeper agent the whole time, and actually worked for a group called Leviathan.

In fairness I had to look all this up, I wasn't particularly familiar with the character beforehand. I just knew enough to know she wasn't the "real" Madame Hydra, who I was more familiar with due to her connection to Wolverine.
